Sequel™ is a company based in Manchester, N.H., working to bring our first product, the twiist™ Automated Insulin Delivery (AID) System powered by Tidepool, to market. What is twiist?
The twiist AID System is a user-friendly automated insulin delivery system designed to provide personalized diabetes management. At its core is the twiist Loop algorithm, which was initially designed by people with diabetes for people with diabetes. The twiist System offers customizable target glucose ranges from 87-180 mg/dL, advanced occlusion detection, and full control from your iPhone and connected Apple Watch. With flexible wear options, 300-unit capacity and a small, discreet form factor, twiist is built for people with diabetes who want to customize their diabetes management to meet their unique needs.
What are some key features of the twiist pump?
The twiist Pump is small but mighty! It weighs less than 2oz, and holds 300 units of insulin, making it the smallest available insulin pump with such a large capacity. It can detect blockages up to 9x faster than other AID systems2
Why did you call it "twiist"?
It is called twiist™ because you twist it to open the device (kind of like an Oreo cookie)
Why are there two i's in twiist?
The ii in twiist stands for intelligent insulin.
Where is twiist manufactured?
The twiist™ Pump is proudly manufactured in the United States.
What is the iiSure™ Technology in your product?
The patented iiSure Technology is an innovative technology that directly measures the volume of insulin delivered and includes 4 checkpoints along the way to help ensure that users get the insulin they expect. This technology also alerts users to blockages (occlusions) up to nine times faster than other insulin pump systems2.
What is the difference between the twiist AID System algorithm and other systems’ algorithms?
- twiist™ has a wider Correction Range than any other commercially available system (as low as 87 mg/dL and as high as 180 mg/dL).1,3-7
- The Correction Range settings can be programmed with up to 48 different segments.
- No other system has a "Premeal Preset" that can be used to target a lower glucose as a person is preparing to eat – this allows for some additional insulin to work prior to eating
- Workout Preset range (as low as 87 mg/dL and as high as 250 mg/dL)
- Emojis to represent carb absorption time (user can see "carbs on board")
- Users may retroactively add or edit the carbohydrate amount entered
- Shows the user the glucose prediction graph that updates every 5 minutes as the algorithm addresses glucose excursions.

Tidepool is a non-profit organization focused on diabetes. This technology is community-driven, designed by and for people living with diabetes, and offers extensive customizable options to truly personalize diabetes management. Why don’t you capitalize the t in twiist?
We tried capitalizing the ‘T’ in twiist, but it just didn’t feel right—like wearing socks with sandals. Plus, lowercase ‘t’ keeps things small, sleek, and stylish… just like our pump!
Is twiist running the latest version of the loop Open Source/DIY algorithm?
The twiist™ AID System is running a modified version of the Tidepool Loop algorithm based on the Open Source/DIY version of Loop. twiist Loop has been cleared by the FDA.
My HCP wants my pump data. How do I do that?
You can share your twiist AID System data with your healthcare provider using the Tidepool Data Platform linked through your My twiist Portal account.
What options for controllers are available with the twiist™ AID System?
The twiist™ App requires an iPhone currently. An Android version is currently in development. The twiist App is also compatible with all released versions of the Apple Watch and Apple Watch OS 4.1 and newer. – This optional component provides a discreet way to view your sensor glucose and insulin information. When you choose to have Loop on, you can also enter carbs, deliver a bolus and more from your Apple watch.
What are the details of the twiist Loop algorithm?
twiist™ AID System powered by Tidepool is designed to help manage your diabetes by adjusting your basal insulin based on a 6-hour prediction of your CGM glucose levels. You can set your correction range between 87-180 mg/dL, and the system will work to keep you in that range. For meals, simply enter the total grams of carbs into the bolus calculator, and it will help you calculate the right insulin dose. You can also give correction doses whenever needed. Plus, you can bolus right from your connected Apple Watch, your iphone, or from the pump itself.
How do you charge the pump? Does it take batteries?
The twiist™ Starter Kit comes with 4 rechargeable batteries and 2 chargers, this helps ensure you always have a battery charged and ready to go.
How long does a battery last?
A fully charged pump battery will power the pump for 3 days (72 hours) and should be swapped each time a cassette (reservoir) change is completed.
How can you wear your twiist Pump?
The twiist™ Pump offers a variety of flexible wear options. It can be secured to a belt or clothing using the provided pump clip, we are designing adhesive patches that will allow it to be worn on body, or it can also be discreetly placed in a clothing pocket like a traditional tubed insulin pump.
Can I bolus from the pump itself?
Yes, you can use the Bolus button to bolus directly from the pump itself, even when you are not close to your phone.
What is the correction/target range?
The Correction Range can be set as low as 87 mg/dL (4.8 mmol/L) and as high as 180 mg/dL (10 mmol/L). It can be set to different values at different times of the day to meet your unique needs. Your Correction Range should be set with the direction of your healthcare provider.
How accurate is twiist in detecting insulin delivery blockage?
twiist features iiSure™ Technology, which provides the fastest blockage detection - up to nine times faster than other AID systems2. This gives you more time to make necessary adjustments to potentially prevent high glucose levels related to infusion set failure.
How many units of insulin will the twiist Pump hold?
The twiist Pump can hold up to 300 units of insulin.
How will users see reports from their pump data?
We are pleased to share that we have partnered with Tidepool. twiist™ users and their doctors will have access to their reports in the Tidepool Data Management System.
Does the twiist AID System algorithm learn with time?
The twiist™ AID System uses the twiist Loop algorithm which uses a 6-hour prediction based on your personal settings, your current glucose value, how much insulin you have on board, the past 30 minutes of blood glucose changes, and your carbohydrate information to adjust every 5 minutes to help keep you in your Target Range. It does not learn with time and only adapts based on the parameters described above.
Does the twiist AID System kick you out of "closed loop" mode?
The twiist™ AID System uses your preferences for whether closed loop mode is on or off. It will keep "looping" if you have CGM values available when Loop is “on”. Otherwise. it will return to your pre-programmed basal settings until your CGM values are available.
Can I use the system in Open Loop mode?
Yes, you can always switch the twiist™ AID System to Loop Off whenever you choose.
Is there a "follow" or "Caregiver" App for the twiist AID System?
Yes, we know how important the caregiver's role is. We have designed the twiist insiight App for them to remotely see key diabetes management data updates, including glucose levels, active insulin, and carbohydrate entries from the twiist AID System. Please note that the insiight App is not intended to be a replacement for the information displayed by the twiist System App and should not inform dosing decisions.

What happens if my phone dies or I lose Bluetooth connection?
When the twiist pump and app are not in communication or your iPhone is powered off, the following functionality is still available:
When Loop is on, the twiist AID System will continue to adjust basal delivery based on information from your CGM.
When Loop is off, the twiist AID System will continue to deliver the programmed basal insulin.
One-button boluses can still be delivered (when enabled in Settings).
Alarm, urgent alert, CGM alert, CGM urgent alert, and alert settings are saved in a manner that is unaffected by power loss.
When and where will the twiist AID System be available?
twiist is currently available in select areas, and we're gradually expanding launch with the goal of providing full national availability before year end. Our phased approach helps us ensure that everyone receives the support they need, from training to user support and ensures Healthcare Provider readiness. Will the twiist Pump be available through the DME channel?
To make the twiist™ AID System the best access experience for providers and people with diabetes, we are focusing on making it available exclusively through the pharmacy channel. We are not currently planning to make the twiist AID System available through the DME channel.
What is the anticipated monthly cost?
Most people will pay $0 the 1st month for the twiist™ AID System and no more than $50 each month thereafter.*
International availability? Launching outside U.S.?
We're working hard to bring the twiist™ AID System to market, starting with our U.S. launch. Our ultimate goal is to make this innovative technology available to people worldwide. We're already taking steps to expand twiist availability beyond the U.S. Regulatory approval and reimbursement vary by region; therefore, we don't have a timeline for international availability yet. Will it be covered under Medicare or Medicaid?

Is there a contract commitment to sign up for twiist?
There is no contract obligation. People living with type 1 diabetes can try twiist without the four-year commitment required by many traditional AID systems, but with the flexibility to manage their diabetes on their terms. Additionally, if you are using an AID system that is in existing warranty, you do not need to wait until your current warranty is up to try twiist.
Do I have to wait until my current warranty is up to try twiist?
People with type 1 diabetes with an AID system in existing warranty can still try twiist through the pharmacy channel. They do not need to wait until their current warranty is up to try twiist.
Who is the twiist AID System cleared for?
The twiist™ AID System is cleared for people 6 and up with type 1 diabetes, with a prescription
What insulin will the twiist AID System be compatible with?
The twiist™ AID System is currently cleared for use with the following fast-acting U-100 insulin: Humalog® and Novolog®.
Can I switch from MDI?
Switching from multiple daily injections (MDI) to the twiist™ AID System could be a game-changer! With MDI, you're used to giving yourself insulin injections multiple times a day. The twiist™ AID System, powered by Tidepool, means no more injections! It automates insulin delivery for you, adjusting your basal insulin based on your real-time sensor data to help keep your glucose levels within your correction range.

Any plans to support control from Android devices?
Our team is working towards having Android compatibility available in the future.
Is a receiver option available if someone does not have a phone?
Currently, the system is designed to work with compatible iPhones only. We are working towards Android phone compatibility in the future.
What kind of training will be available to people living with diabetes?
Sequel™ will offer in-person training by Certified twiist™ Trainers, ensuring you get the support you need to use the twiist AID System confidently and effectively.
What kind of Customer Support will you have? Will it be 24/7? What languages will you support?
We will have 24/7 live product support with representatives fluent in both English and Spanish. In addition, you’ll have access to Diabetes Educators and a dedicated Customer Care Team to support you throughout your journey using the twiist™ AID System.

Is it hard to set up?
The setup is similar to traditional insulin pumps. Healthcare providers (HCPs) will assist people with diabetes with determining their Correction Range and other insulin pump settings.
People with diabetes need only enter their carbohydrates and emojis representing the type of food, and maintain the system
Do you have to build the App yourself?
Unlike traditional Open Source or DIY Loop, you are not required to build the twiist™ App yourself. twiist Loop is provided as part of the twiist AID system.
Why did you choose the Abbott FreeStyle Libre 3 Plus sensor?
We chose the Libre 3 Plus sensor as our first CGM partner because it is the world’s smallest sensor *1, has proven accuracy3 for up to 15 days, and is easy to use2 .

Is Sequel planning to add additional CGM partners?
Yes, we recently announced the twiist™ AID System will become the first AID system compatible with the Senseonics Eversense 365, the world’s first and only one-year CGM system. We will be looking to add additional CGM partners in the future in order to make advanced diabetes management more accessible and flexible for people living with diabetes. We are on a mission to improve the lives of people with diabetes and give them choice in how they manage their diabetes. Having multiple CGM options, so people can select the one that is best for them, is one of the ways we will bring this to life.
